Selecting the Right Pore Size

The pore size refers to the diameter of the openings within the membrane and to the effectiveness in filtering out particles of a certain size. Choosing the right membrane pore size depends on the level of sterility or clarification required, as well as the process speed needed for your application.

0.22µm (Sterilizing Grade): 

This is the standard choice for sterilization when working with tissue culture media, buffers, or biological fluids, and is designed to filter virtually all bacteria and microorganisms. The flow rate is slower and controlled.

Applications include:

  • Cell culture media and buffers

  • Microbiological testing

  • Sensitive analytical workflows

0.45µm (Clarification Grade): 

This size is used for general clarification, and it is ideal for viscous samples, larger volumes, and removing larger particulates, yeast, and mold. It allows for faster flow rates, lower resistance, and it is a good fit if complete sterility is not required. 

Applications include:

  • Buffer and solvent filtration

  • Environmental or biological sample clarification

  • Pre-filtration before finer membranes

Additionally, if working with particle-heavy samples, as a first step, a 0.45µm filter can be used before the 0.22µm filter to help reduce the risk of membrane clogging and improve the flow rates. 

The Streamlined Setup Process

If you choose a vacuum filtration system that you attach to a flask or bottle, the setup is quick and easy. 

  1. Secure the Filter: By screwing the filter directly onto the threaded neck of your media bottle or reusable receiver bottle to create an airtight seal for collecting sterile filtrate.

  2. Connect the Vacuum: Attach the vacuum hose to the side arm of the filtration unit, and the other end to your vacuum pump.

  3. Create the Seal: Make sure the connections are secured tightly to maintain the airtight system, and a consistent vacuum pressure is essential to ensure proper filtration speed.

  4. Operation: It’s important to pour your sample into the funnel reservoir before switching on. Once the vacuum is applied, negative pressure will draw the liquid through the membrane into the collection bottle below.

  5. Filter: Once finished, the collection will be in the bottle below. Turn off the vacuum and slowly release the pressure, disconnecting the tubing and sealing to maintain sterility.

Vacuum Filtration vs. Gravity Filtration

Compared to gravity filtration, which relies on slow natural flow, vacuum filtration systems can be more efficient by using negative pressure to push the liquid through the membrane.
